چکیده

ABSTRACT: The molecular background of canine mast cell tumors (MCT) has been extensively investigated; however, the dynamic changes that occur during carcinogenesis and metastasis are not fully understood. This study aimed to evaluate incidence mutations in c-KIT proto-oncogene MCTs relative draining regional lymph nodes. Suspected or confirmed node was classified accordingly HN Weishaar classification. included 34 dogs diagnosed with MCT; 19 patients were enrolled prospectively. These had primary MCT resected analyzed simultaneously. second group evaluated retrospectively fifteen resectioning without evaluation node. Analyzes mutation performed for all and, first group, compared between HN-classified metastasis. Internal tandem duplications (ITD) exon 11 gene detected 20% patients. Ten nineteen (52%) presented infiltration node, ITD five two from Groups 1 2, respectively. common may be found metastases/mast infiltrates absence tumor. Evaluation tumor metastases informative defining both prognosis therapeutic options cases.

Imatinib Responsiveness in Canine Mast Cell Tumors Carrying Novel Mutations of c-KIT Exon 11

In 2 individual cases of canine mast cell tumors, we identified 2 novel c-KIT mutations in exon 11: a 9-base pair (bp) deletion (c.1663-1671del) and a point mutation (c.1676T>A). The 9-bp deletion mutation caused a loss of 3 amino acids, corresponding to p.Gln555_Lys557del, and the point mutation resulted in the substitution of valine by aspartic acid (p.Val559Asp) in the juxtamembrane domain o...

Mutations in C-KIT exon 11 in canine cutaneous mast cell tumors

Background The c-KIT proto-oncogene encodes the receptor tyrosine kinase KIT, which has been shown to play important roles in the cellular maturation, survival, proliferation, and migration of several cell types including mast cells. Mast cell tumors (MCTs) are the most common cutaneous tumor in the dog. MCTs exhibit wide variation in biological behavior. KIT mutations and aberrant KIT expressi...

Canine Mast Cell Tumors

Initial work up of a mast cell tumor (MCT)will involve diagnosis by FNA and staging. The initial staging performed will depend on the index of suspicion that the mass is a grade II or III MCT and clinician’s preference. Cytology is generally diagnostic for MCT. However, an incisional biopsy to determine grade is recommended in some cases. The histological grade may be important to determine in ...

Mast Cell Leukaemia: c-KIT Mutations Are Not Always Positive

Mast cell leukemia (MCL) is a rare and aggressive disease with poor prognosis and short survival time. D816V c-KIT mutation is the most frequent molecular abnormality and plays a crucial role in the pathogenesis and development of the disease. Thus, comprehensive diagnostic investigations and molecular studies should be carefully carried out to facilitate the therapeutic choice. A MCL patient's...
